采用RDP试剂(改进自ChomczynskiandSacchi1987)提取锯缘青蟹精巢和卵巢的总RNA,经oligotex试剂盒纯化得到mRNA.利用SMART(switchingmechanismat5'endofRNAtranscript)技术,构建成精巢和卵巢的全长cDNA文库.在构建好的文库中,精巢的文库含有约1.45×106的重组子,卵巢的文库含有约为1.75×106的重组子.插入cDNA长度为500bp~3kb,说明两文库均具有良好的质量,为进一步筛选、克隆精巢与卵巢特异表达基因奠定了基础.

应用特征光合色素研究东海和南海北部浮游植物的群落结构 - 陈纪新; 黄邦钦; 刘媛; 曹振锐; 洪华生
应用高效液相色谱并结合二极管阵列检测器分析技术,研究了东海与南海北部典型断面浮游植物的光合色素组成,进而由CHEMTAX软件估算浮游植物群落结构。结果表明:东海断面浮游植物叶绿素a生物量大于南海北部断面。受海水层化结构影响,东海PN断面浮游植物群落结构形成上层和下层两种类型,上层以蓝藻为主要优势类群;下层以硅藻为主要优势类群。南海北部S断面浮游植物群落形成近岸与离岸两种类型,近岸以硅藻、隐藻、绿藻为主要优势类群;离岸以定鞭金藻、蓝藻、原绿球藻为主要优势类群。初步分析了研究海区浮游植物群落结构与环境因子的关系。

本文对TOA萃取剂萃取钍适宜的操作条件及萃取溶剂进行了试验,提出了TOA/苯萃取234Th的最佳条件: 在 1cm3含 234Th的 6mol/dm3 HNO3 溶液中,用体积比为 0. 5的TOA/苯溶液 2cm3 萃取,然后用 2cm3 1. 0mol/dm3 H2SO4反萃取,最高萃取率达 94%.对TOA/苯、TBP/煤油和TTA/苯这 3种萃取体系的萃取效率及其影响因素、萃取机理、成本等做了比较,认为 3种体系各有利弊,但均可有效地应用于相关的海洋学研究中.
