  1. Phosphonocarboxylates inhibit the second geranylgeranyl addition by Rab geranylgeranyl transferase

    Rab geranylgeranyl transferase (RGGT) catalyzes the post-translational geranylgeranyl (GG) modification of (usually) two C-terminal cysteines in Rab GTPases. Here we studied the mechanism of the Rab geranylgeranylation reaction by bisphosphonate analogs in which one phosphonate group is replaced by a carboxylate (phosphonocarboxylate, PC). The phosphonocarboxylates used were 3-PEHPC, which was previously reported, and2-hydroxy-3-imidazo[1,2-a] pyridin-3-yl-2-phosphonopropionic acid ((+)-3-IPEHPC), a >25-fold more potent related compound as measured by both IC50 and Ki. (+)-3-IPEHPC behaves as a mixed-type inhibitor with respect to GG pyrophosphate ( GGPP) and an uncompetitive inhibitor with respect to Rab substrates.   2. Rab27a and Rab27b control different steps of the exosome secretion pathway

    Exosomes are secreted membrane vesicles that share structural and biochemical characteristics with intraluminal vesicles of multivesicular endosomes (MVEs). Exosomes could be involved in intercellular communication and in the pathogenesis of infectious and degenerative diseases. The molecular mechanisms of exosome biogenesis and secretion are, however, poorly understood. Using an RNA interference (RNAi) screen, we identified five Rab GTPases that promote exosome secretion in HeLa cells.     Melanosomes, the pigment granules that provide tissues with colour and photoprotection, are the cellular site of synthesis, storage and transport of melanin pigments. They are synthesised in mammalian skin melanocytes, in choroidal melanocytes and retinal pigment epithelial (RPE) cells in the eye, and in melanophores (a class of pigment-containing cells) in lower vertebrates.
