  1. Effect of the chronic treatment with aqueous extract of Caesalpinia ferrea and Chrysobalanus icaco on the vascular reactivity of diabetic rats

    Caesalpinia ferrea and Chrysobalanus icaco are plants commonly used by the folk medicine for the treatment of diabetes mellitus. In this work, the effects of the chronic treatment with aqueous extracts of C. ferrea (AECF) and C. icaco (AECI) on the vascular reactivity of alloxan-induced diabetic rats were evaluated. In rings of mesenteric artery, AECF and AECI were not able to modify the contractions induced by phenylephrine or relaxations induced by carbachol.   2. Determination of dextromethorphan in rabbit plasma by LC-MS/MS and its application to pharmacokinetic study

    A highly sensitive liquid chromatographic tandem mass spectrometric (LC-MS/MS) method for determination of dextromethorphan in rabbit plasma using triazolam as the internal standard (IS) was developed. Plasma samples were extracted with ethyl acetate and separated on a SB-C18 column with a mobile phase of acetonitrile-water 60:40 (v/v) at a flow of 0.3 mL/min.   3. Quantificação de ácido (-) hidroxicítrico em extratos comerciais de Garcinia cambogia por cromatografia líquida de alta eficiência

    O extrato de Garcinia cambogia (GC), utilizado no tratamento da obesidade, é padronizado por quantificação de ácido hidroxicítrico (AHC) em sua forma salina (CaAHC). Assim, objetivou-se quantificar AHC em duas amostras comerciais de GC (A e B) com 50 % de AHC, de acordo com os laudos de origem. O rendimento extrativo por maceração foi 23,83 % (A) e 22,37 % (B).     The objective was to develop and validate a method for the determination of total polyphenols in a vaginal gel containing a dried extract of Aroeira (Schinus terebinthifolius Raddi). The method was developed by UV spectrophotometry, and proved to be precise, accurate, reproducible, and easy to perform.

  5. Solubility of pioglitazone hydrochloride in polyethylene glycol 600-ethanol-water mixtures at 25 °C

    The solubility of pioglitazone hydrochloride in polyethylene glycol 600-ethanol-water mixtures at 25 °C was determined using flask shake method. The generated data extended the solubility database for further computational investigations and also was used to assess the prediction capability of a trained version of the Jouyban-Acree model for solubility prediction in mixed solvents at various temperatures. The accuracy of the predicted solubilities was evaluated by the mean percentage deviation (MPD) between the predicted and experimental solubilities. The overall MPD of the Jouyban-Acree model for the back-calculated solubility data in binary and ternary solvent mixtures was 13.1 ± 12.9 %

    The present work was to study the anti-inflammatory activity of Coccinia indica Wight and Arn fruits belonging to family Cucurbitaceae. The fruit powder of C. indica was subjected to successive extraction with petroleum ether, chloroform, ethanol and water in a Soxhlet extractor. The ethanol extract after preliminary phytochemical investigation shown the presence of glycosides, triterpenoids, flavonoids, tannins and phenolic compounds.     Se ensayó la actividad antimicobacteriana in vitro de los extractos diclorometánico (DCM) y metanólico (MeOH) de G. cabrerae y G. megapotamica var. scabiosoides frente a dos cepas de Mycobacterium tuberculosis (H37Rv y MDR), por medio del test colorimétrico TEMA, determinándose la concentración inhibitoria minima (MIC) en ug extracto/ml.   8. Desenvolvimento e validação de metodologia analítica por cromatografia líquida de alta eficiência para determinação de paracetamol em comprimidos

    Um método simples e sensível por CLAE foi desenvolvido para a determinação quantitativa de paracetamol em comprimidos. Condições cromatográficas para uma eficiente separação foram obtidas empregando-se fase estacionária C8, fase móvel de KH2 PO4 40 mM pH 6,0 e metanol 85:15 (v/v), com fluxo de 1,2 mL/min. A análise de regressão mostrou um coeficiente de correlação superior a 0,99. O método apresentou recuperação consistente para paracetamol (97,03-100,57 %). A precisão intra-dia e precisão intermediária não ultrapassou 1,87 e 0,95 % do CV, respectivamente.   9. Antinociceptive activity of the leaf essential oil of Coleus aromaticus Benth

    The present study was undertaken to evaluate the possible antinociceptive effect of Coleus aromaticus Benth oil. The pain sensitivity was assessed by the acetic acid induced writhing and hotplate models in mice. The essential oil exhibited a significant reduction in acetic acid induced writhing response, a dose dependent increase in reaction time against thermal pain and a moderate motor impairment at analgesic doses and significant reduction in endurance time at higher dose on rotarod apparatus.   10. Caracterização físico-química do tripanomicida benznidazol para o desenvolvimento de medicamentos

    Este trabalho objetivou a caracterização físico-química do benznidazol (BNZ), utilizando diversas técnicas analíticas. Os dados obtidos nos espectrogramas de infravermelho mostram as características principais da carbonila e deformação N-H; as vibrações devido ao grupo benzílico e imidazólico. O BNZ apresentou pico endotérmico em aproximadamente 190 ºC referente a fusão. O perfil difractométrico revela a presença de picos de maior intensidade a 7,5; 16,5 e 22 º2θ, evidenciando o comportamento cristalino do BNZ, corroborando com as micrografias obtidas.   11. Required Hydrophilic-Lipophilic Balance values of octyldodecanol from emulsion stability tests and relative dielectric permittivity measurements

    Although octyldodecanol has been used for several decades as an ingredient for emulsions, no required Hydrophilic-Lipophilic Balance (HLB) values for this alcohol were found in the literature. The purpose of this paper is to present the results of the experimental determination of the required HLB values, from emulsion stability tests and from low-frequency relative dielectric permittivity measurements. The required HLB values obtained from emulsion stability tests for oil-in-water and water-in-oil emulsions are 11.2 and 5.9, respectively. By relative dielectric permittivity measurements, the required HLB value obtained for oil-in-water emulsions was 10.9 ± 0.4, in agreement with the results from emulsion stability tests.

  12. Development and validation of an analytical method for the simultaneous determination of benzoylecgonine and ecgonine methyl ester in human urine by gas chromatography/mass spectrometry

    A simple and rapid procedure has been developed and validated for the simultaneous determination of benzoylecgonine and ecgonine methyl ester in human urine samples. After extraction and sample cleanup by solid-phase extraction (SPE), the extracts were derivatized with 50 uL of MTBSTFA and analyzed by gas chromatography/mass spectrometry (GC/MS). The limit of detection (LOD) was 50 ng/mL for both metabolites. The recoveries were 81% and 75.3% for benzoylecgonine and ecgonine methyl ester, respectively.   14. Antioxidant activity and acute oral toxicity of Phrygilanthus acutifolius flowers

    Antioxidant activity of ethanol and water extracts and infusion of the flowers of Phrygilanthus acutifolius were evaluated. Total phenolic contents were determined using Folin- Ciocalteau reagent. The ethanol extract was more effective in scavenging radical 1,1 biphenyl -2-picrylhydrazyl (DPPH), with lower IC50 values. The water and ethanol extracts were the most effective antioxidants based on the inhibition of lipid peroxidation (β carotene bleaching method).   15. In vivo studies on the bronchodilatory and analgesic activities of Hyoscyamus niger and Aspalathus linearis

    The present study describes the in vivo bronchodilatory and analgesic activities of the crude extract of Hyoscyamus niger (Hn.Cr) and Aspalathus linearis (Al.Cr), which tested positive for alkaloids, coumarins, flavonoids, sterols, tannins, terpenes and flavonoids, tannins, saponins respectively. Hn.Cr (0.3-3.0 mg/kg) and Al.Cr (10-100 mg/kg) caused inhibition of the CCh (1 μmol/kg)-induced increase in the inspiratory pressure of anaesthetized rats. Hn.Cr and Al.Cr dose-dependently (50-100 mg/kg) reduced the numbers of acetic acid-mediated writhes in mice. These results indicate that Hyoscyamus niger and Aspalathus linearis exhibit airways-relaxant and anti-nociceptive effects, hence justifying their medicinal use in asthma and pain.

    Transdermal drug delivery systems of venlafaxine hydrochloride were prepared by using combination of hydrophilic (HPMC E15) and hydrophobic (ERS100 and ERL 100) polymers in 1:5, 2:4, 3:3, 4:2, 5:1 ratios by solvent casting technique with 15 % v/w propylene glycol as plasticizer.     The present study involves development and investigation of liposomal system for improving skin residence of ciclopirox olamine in cutaneous mycosis. Spherical unilamellar liposomes of ciclopirox olamine were prepared by ethanol injection method. The vesicle size and % entrapment efficiency were in the range of 196 ± 1.73 to 1040.66 ± 7.02 nm and 34.28 ± 4.4 to 54.89 ± 1.9 respectively.   20. Vacuum foam dried sugar-phosphate amorphous mixtures for stabilization of doxorubicin hydrochloride

    The objective of the present study was to stabilize doxorubicin hydrochloride in sugar-phosphate amorphous mixtures at ambient temperature by drying it with vacuum foam drying. Finished products were evaluated for foam characteristic, residual moisture content, reconstitution time, percent drug recovery and drug-excipient interactions. FTIR studies revealed existence of physical interaction of drug with sugar. Light microscopy showed formation of amorphous glass which was supported by the observations of XRPD analyses. The optimized composition in vacuum foam drying was processed by lyophilization and their stability was compared.
