  1. Potassium Starvation in Yeast: Mechanisms of Homeostasis Revealed by Mathematical Modeling

    The intrinsic ability of cells to adapt to a wide range of environmental conditions is a fundamental process required for survival. Potassium is the most abundant cation in living cells and is required for essential cellular processes, including the regulation of cell volume, pH and protein synthesis. Yeast cells can grow from low micromolar to molar potassium concentrations and utilize sophisticated control mechanisms to keep the internal potassium concentration in a viable range. We developed a mathematical model for Saccharomyces cerevisiae to explore the complex interplay between biophysical forces and molecular regulation facilitating potassium homeostasis.   2. Adaptation to potassium starvation of wild-type and K+-transport mutant (trk1,2) of Saccharomyces cerevisiae: 2-dimensional gel electrophoresis-based proteomic approach

    Saccharomyces cerevisiaewild-type (BY4741) and the correspondingmutant lacking the plasma membrane main potassium uptake systems (trk1,trk2) were used to analyze the consequences of K+ starvation following a proteomic approach. In order to trigger high-affinity mode of potassium transport, cells were transferred to potassium-freemedium.Protein profilewas followed by two-dimensional (2-D) gels in samples taken at 0, 30, 60, 120, 180, and 300 min during starvation.We observed a general decrease of protein content during starvation that was especially drastic in the mutant strain as it was the case of an important number of proteins involved in glycolysis.   13. Investigación de anticuerpos incompletos o bloqueantes en el diagnóstico de la brucelosis caprina

    Se estudian cien sueros caprinos procedentes de rebaños brucelósicos. Sometidos a la prueba de seroaglutinación lenta, resulta negativos el 73,00 p.100. Realizada la prueba de Coombs, con éstos últimos, se connsigue un 69,86 p.100 de resultados positivos, lo que demuestra la mayor sensibilidad de este método en la detección de anticuerpos antibrucela.

    A serologic study has been carried out of 20 serum samples from mares with different pathologic process (abortions and poll-evil). The 60.000 p.100 of the adults ,mares, show positive title in agglutination test with B. abortus antigen. Only one serum was positive with B. melitensis antigen. Through complement fixation test it was detected only one positive serum (mare with poll-evil). A B. abortus strain was isolated from exudate: of poll-evil.

    It has been done an in vitro study about the antibacterial activity of Cefamandole with 459 bacterial strains of different origin. Wi have found a good sensibility to the antibiotic for the Brucella (B. abortus), Enterobacteriaceae, Staphylococcus listeria (L. monocitogenes) and E. rhusio? pathiae.

  16. Determinación de enterobacteriáceas en raciones experimentales y piensos compuestos para animales domésticos

    It has been done a bacteriological screening to detect enterobacteriaceac in 50 samples of experimental rations and commercial nixed for domestic animals. We have found that 36 samples (72 p. 100) were contamined with enterobacteries. The limits of contamination were between 200 - 15 x .106 organisms/g. The strains isolated belong to the species: E. hafniae (48,42 p. 100), E, cloacae (28,42 o. 100), E. aerogenes (9,47 p. 100), E. liquefaciens (2,11 p. 100), E. herbicola (4,21 p. 100), E. quercina (1,05 p. 100) y E. coli (632 p. 100). These last ones had the serotypes 0.18:K76:14, 0,26:K60-11,044:K24:18 and 055...

    From combs, with typical characters of European foulbrood it has been found a strain of B. alvei with reducer capacity for nitrates and growing in Saboureaudglucose medium.

  18. Estudio comparativo entre el método estandar y la técnica mejorada con SMCA para la detección y recuento de bacterias proteolíticas

    For the detection and count of proteolytic bacteria in 83 samples of raw milk we have used the standard method (A.P.H.A., 1967) and the improved technics by Martley et at, 1970, with SMCA (Standard Method Caseinate Agar). In the plates with SMCA the count was always higher than in the standard method (29 p. 100 as the total average) and the reading in that medium was easier and quicker because after 12-18 h. of incubation it was possible to detect more than p. 100 of the proteolytic bacteria.

    For the study the 150 staphylococcal strains —of human and animal origin¬it has been carried out the tests of mannitol (aerobic and anaerobic), DN-ase, coagulase,hemolysis and proteolytic activity on casein and gelatin. Throughout the work it has been seen that there is a significative difference between the proteolytic capacity of the DN-ase positive and DN-ase negative staphylococci. So, this evidence can be an early and interesting complementary test for the staphylococcal identification.

    A comparison was made between the bacteriological quality grades, given to the same milk samples by the Standard Plate Count (SPC), the Methylene Blue Reduction Fest (MBRT) and the Resazurine Reduction Test (RRT); both the 10 minutes test and the time taken to reach a 2,5 value at the Lovibond "1.000" were applied to the RRT. A close relation was found between the SPS MBRT (r =-0,82); a similar relation was observed between the SPC and RRT (r= -0,88) and so between the SPC and the time to reach a 2,5 value at the comparator (r = -0,84).
