  1. Characterization of multidrug-resistant diabetic foot ulcer enterococci

Semedo-Lemsaddek, Teresa; Mottola, Carla; Alves-Barroco, Cynthia; Cavaco-Silva, Patrícia; Tavares, Luís; Oliveira, Manuela
Introduction: Diabetes mellitus is a highly prevalent chronic progressive disease with complications that include diabetic-foot ulcers. Methods: Enterococci isolated from diabetic-foot infections were identified, evaluated by macrorestriction analysis, and screened for virulence traits and antimicrobial resistance. Results: All isolates were considered multidrug-resistant, cytolysin and gelatinase producers, and the majority also demonstrated the ability to produce biofilms. Conclusions: These results indicate the importance of enterococci in diabetic-foot infection development and persistence, especially regarding their biofilm-forming ability and resistance to clinically relevant antibiotics.

  4. In vitro design of a novel lytic bacteriophage cocktail with therapeutic potential against organisms causing diabetic foot infections

Mendes, João J.; Leandro, Clara; Mottola, Carla; Barbosa, Raquel; Silva, Filipa A.; Oliveira, Manuela; Vilela, Cristina L.; Melo-Cristino, José; Górski, Andrzej; Pimentel, Madalena; São-José, Carlos; Cavaco-Silva, Patrícia; Garcia, Miguel
In patients with diabetes mellitus, foot infections pose a significant risk. These are complex infections commonly caused by Staphylococcus aureus, Pseudomonas aeruginosa and Acinetobacter baumannii, all of which are potentially susceptible to bacteriophages. Here, we characterized five bacteriophages that we had determined previously to have antimicrobial and wound-healing potential in chronic S. aureus, P. aeruginosa and A. baumannii infections. Morphological and genetic features indicated that the bacteriophages were lytic members of the family Myoviridae or Podoviridae and did not harbour any known bacterial virulence genes. Combinations of the bacteriophages had broad host ranges for the different target bacterial species. The activity...

  6. Histomorphometric evaluation of the small coronary arteries in rats exposed to industrial noise

Lousinha, Ana; Antunes, Eduardo; Borrecho, Gonçalo; Oliveira, Maria João; Brito, José; Martins dos Santos, José
Morphological changes induced by industrial noise (IN) have been experimentally observed in several organs. Histological observations of the coronary arteries showed prominent perivascular tissue and fibrosis among IN-exposed rats. The effects on the small arteries are unknown. Objective: To evaluate the histomorphometric changes induced by IN on rat heart small arteries. Methods: Twenty Wistar rats exposed to IN during a maximum period of seven months and 20 age-matched controls were studied. Hearts were transversely sectioned from ventricular apex to atria and a mid-ventricular fragment was selected for analysis. The histological images were obtained with an optical microscope using 400× magnifications. A total of 634 arterial vessels (298 IN-exposed and...

  7. Feasibility of wavelength dispersive X-ray fluorescence spectrometryfor the determination of metal impurities in pharmaceutical productsand dietary supplements in view of regulatory guidelines

Figueiredo, Alexandra; Fernandes, Tânia; Costa, Isabel Margarida; Gonçalves, Luísa; Brito, José
The aim of this study was to investigate the feasibility of Wavelength Dispersive X-ray Fluorescence (WDXRF) spectrometry for the measurement of As, Cd, Cr, Cu, Hg, Ir, Mn, Mo, Ni, Os, Pb, Pd, Pt, Rh, Ru and V impurities in pharmaceuticals and dietary supplements, in view of the requirements by EMA and USP for the measurement of elemental impurities in drug products and according to the International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use (ICH guidelines). For that purpose, a 4 kW WDXRF spectrometer (S4 Pioneer, Bruker AXS) was used after system calibration. The...

  8. Arthritis induces early bone high turnover, structural degradation and mechanical weakness

Vidal, Bruno; Cascão, Rita; Vale, Ana Catarina; Cavaleiro, Inês; Vaz, Maria Fátima; Brito, José Américo Almeida; Canhão, Helena; Fonseca, João Eurico
BACKGROUND: We have previously found in the chronic SKG mouse model of arthritis that long standing (5 and 8 months) inflammation directly leads to high collagen bone turnover, disorganization of the collagen network, disturbed bone microstructure and degradation of bone biomechanical properties. The main goal of the present work was to study the effects of the first days of the inflammatory process on the microarchitecture and mechanical properties of bone. METHODS: Twenty eight Wistar adjuvant-induced arthritis (AIA) rats were monitored during 22 days after disease induction for the inflammatory score, ankle perimeter and body weight. Healthy non-arthritic rats were used as controls for...

  9. Analysis of micronutrients and heavy metals in portuguese infant milk powders by wavelength dispersive x-ray fluorescence spectrometry (WDXRF)

    Fernandes, Tânia A. P.; Brito, José A. A.; Gonçalves, Luísa M. L.
Concentrations of trace mineral nutrients such as Cu, Fe, and Zn have been analyzed by WDXRF in eight different infant milk powders, representing the majority of the milk powder brands commercialized in Portuguese pharmacies. Results suggest that the concentrations of some of elements analyzed are not consistent with the values labeled by manufacturers. However, all fall inside the concentration ranges defined by the Portuguese and European legislations, except for Al. The presence of aluminum was detected in all analyzed formulations in an average concentration of 8.25 to 19.75 mg Kg−1. This study also reveals the feasibility of WDXRF for the quantitative analysis...

  15. Assessing transmissibility of HIV-1 drug resistance mutations from treated and from drug-naive individuals

Winand, Raf; Theys, Kristof; Eusébio, Mónica; Aerts, Jan; Camacho, Ricardo J.; Gomes, Perpétua; Suchard, Marc A.; Vandamme, Anne-Mieke; Abecasis, Ana B.; on behalf of the Portuguese HIV-1 Resistance Study Group
OBJECTIVES: Surveillance drug resistance mutations (SDRMs) in drug-naive patients are typically used to survey HIV-1-transmitted drug resistance (TDR). We test here how SDRMs in patients failing treatment, the original source of TDR, contribute to assessing TDR, transmissibility and transmission source of SDRMs. DESIGN: This is a retrospective observational study analyzing a Portuguese cohort of HIV-1-infected patients. METHODS: The prevalence of SDRMs to protease inhibitors, nucleoside reverse transcriptase inhibitors (NRTIs) and nonnucleoside reverse transcriptase inhibitors (NNRTIs) in drug-naive and treatment-failing patients was measured for 3554 HIV-1 subtype B patients. Transmission ratio (prevalence in drug-naive/prevalence in treatment-failing patients), average viral load and robust linear regression with outlier...

  16. Discordant predictions of residual activity could impact dolutegravir prescription upon raltegravir failure

Theys, Kristof; Abecasis, Ana; Libina, Pieter; Gomes, Perpétua; Cabanas, Joaquim; Camacho, Ricardo J.; Van Laethem, Kristel
BACKGROUND: Dolutegravir is approved for the treatment of HIV-1 patients exposed to other integrase inhibitors, but the decision to use dolutegravir in this setting should be informed by drug resistance testing. OBJECTIVES: This study determined the extent of disagreement in predicted residual dolutegravir activity after raltegravir use, and identified individual mutational patterns for which uncertainty exists among HIV-1 expert systems. STUDY DESIGN: Mutation patterns were classified in raltegravir signature pathways including positions 143, 148 and 155, and interpreted into clinically informative resistance levels using genotypic drug resistance interpretation systems ANRS v24, HIVdb v7.0 and Rega v9.1.0, and instructions of dolutegravir use as approved by the...
