Pea seed pre hydration and its interference with the evaluation of the physiological potential - Caroline Jácome Costa
The present work had the objective of to study biochemical andphysiological alterations in function of pea seed pre hydration with e phasis in theelectrolyte leakage and in the expression of different isoenzymatic systems,trying tocontribute for the i provement of the pre hydration methods of seeds to be submitted totests for evaluation of its physiological potential.Seeds lots of cvs.Axé and Maria,characterized for the water content,germination,first ger mination counting,acceleratedaging,electrical conductivity and seedling emergence in the field,were used.The seeds ofeach lot were pre hydrated in saturated atmosphere and hum idified substratu until theyreach 10,12 and 14%of water content.After the pre hydration,the seeds were submitted tothe tests of electrical conductivity,accelerated aging and leakage of calcium ,potassium andmagnesium and characterized for the expression pattern of the isoenzymatic systemsMalate Dehydrogenase,Acid Phosphatase,Glutamate Dehydrogenase and GlutamateOxalacetate Transaminase.The pre hydration of the seeds using saturated atmosphere orhumidified substratum see s to affect the process of restructuring the seed cellulare branes system differently,being reflected in alterations in the pattern of electrolyteleakage and Acid Phosphatase isoenzymatic expression.Discrepancies take place a ongresults obtained by the test of electrical conductivity for evaluation of the physiologicalpotential of pea seeds,depending on the proceeding adopted to pre hydrate the seeds.

Monoclonal antibodies against LigANI and LigBNI ofpathogenic leptospires:production and characterization - Núbia Seyffert
Leptospiral immunoglobulin-like (Lig)proteins are exposed on membrane surface ofpathogenic Leptospira and may play a role in host cell attachment and invasion duringinfection.A pair of Ligs,named LigA and LigB,with identical and non-identicalpolypeptide regions has recently been identified.The two proteins elicit a stronghumoral immune response during acute host infection and have been suggested astargets for development of vaccines and diagnostic tests for leptospirosis.This studyreports the production and characterization of monoclonal antibodies (Mabs)againstrecombinant non-identical fragments of LigA (rLigANI)and LigB (rLigBNI).Therecombinant fragments were used for mice immunization and screening hybridomasby indirect ELISA.Four Mabs obtained against rLigANI and six Mabs obtainedagainst rLigBNI were isotyped and evaluated regarding their potential for use instudies of immunoprotection and diagnostic test development.The Mabs were of theIgM (1),Ig 2b (3)and Ig 1 (6)isotypes and reacted with the native proteins from apathogenic strain of Leptospira i terroga s serovar Copenhageni L1 130 in an indirectELISA and immunofluorescence.Mabs affinity constants felt between 4x10 7 M -1 and2x10 8 M -1 ,and epitope mapping by additive ELISA has shown that each Mab eitherreact with the same epitope in the recombinant molecule or cause steric hindrance.Tissue sections from kidneys of hamsters experimentally infected with leptospiresand probed with the Mabs reacted positively by immunohistochemistry.Thesefindings suggest that the Mabs obtained can be useful for the studies ofimmunoprotection and development of diagnostic tests.

Relações entre Índices de Instabilidade eas chuvas ocorridas em Porto Alegre,RS,de 1990 a 2005 - Nélia Kainara Rodrigues Cardoso
O objetivo geral do presente trabalho foi verificar as relações estatísticas,dos índicesde instabilidade com o total de precipitação ocorrida,em faixas de valores.Paradeterminar a confiabilidade de cada um dos índices de instabilidade aqui estudados,pois a precipitação age diretamente na agricultura,no planejamento de festas eeventos,no vestuário,enfim,fazendo com que o estudo desta variável,seja cadavez mais explorado,a fim de que haja um melhor entendimento,e com isso possaser feito um bom planejamento para os exemplos citados acima,e tantos outros.Aprecipitação,bem como sua forma,está associada às características de instabilidadeda atmosfera,essa variável meteorológica é determinada através de estudos daconfiguração da atmosfera superior,utilizando informações de sondagensaerológicas e essas informações são coletadas pelas estações de ar superior,existentes em poucas localidades no Brasil.Na pesquisa proposta,foram usadosapenas índices de instabilidade que envolvam equações matemáticas alimentadaspor valores de variáveis atmosféricas obtidas de sondagens aerológicas.Naexecução desse trabalho serão usados dados diários de precipitação da estaçãometeorológica de Porto Alegre pertencente ao 8 0 Distrito de Meteorologia do InstitutoNacional de Meteorologia .
