25. Metabolic and biochemical responses of probiotic bacteria to oxygen - Talwalkar, Akshat; Kailasapathy, Kasipathy
The interaction between oxygen and probiotic bacteria was studied by growing Lactobacillus acidophilus and Bifidobacterium spp. in 0, 5, 10, 15, and 21% oxygen in a hypoxic glove box. The metabolic responses of each probiotic strain in the different oxygen environments were monitored by measuring the levels of lactic acid and determining the lactate-to-acetate ratio. Biochemical changes induced by oxygen were examined by monitoring the specific activities of NADH oxidase, NADH peroxidase, and superoxide dismutase. In addition, the ability to decompose hydrogen peroxide and the sensitivity of each strain to hydrogen peroxide was also determined. With an increase in oxygen...

29. The isolation, growth and survival of thermophilic bacteria from high temperature petroleum reservoirs - Grassia, Gino Sebastian
The microbial ecology of 45 high temperature (> 50 ° C) petroleum reservoirs was investigated by isolating and characterizing bacteria that were present in their produced fluids. Initial work was aimed at selecting a suitable high temperature petroleum reservoir for the study of natural microbial populations. Experimental work then focussed on establishing the physico-chemical conditions that prevail in the selected reservoir and on developing media and enrichment conditions for the isolation of microorganisms indigenous to the reservoir. The ability of reservoir bacteria to grow and survive under the physical and chemical conditions found in the selected reservoir was used to assess the likelihood of an indigenous origin for...

31. Bacteria isolated from a sugarcane agroecosystem: their potential production of polyhydroxyalcanoates and resistance to antibiotics - Lima,Teresa Cristina S. de; Grisi,Breno M.; Bonato,M. Christina M.
In this investigation, a sugarcane agroecosystem at a coastal tableland, in the northeast of Brazil, was screened to obtain bacteria strains able to synthesize poly-b-hydroxyalkanoates (PHA), using sucrose as the main carbon source. The potential to synthesize PHA was tested qualitatively by Sudan Black staining of colonies growing in different carbon sources: sucrose, glucose, fructose, propionate and cellulose. In a typical sugarcane crop management system, the plantation is burned before harvesting and vinasse, a byproduct of alcohol production, is used in a fertirrigation system causing, probably, selective pressures on the microbiota of natural environments. Eightytwo bacteria strains, belonging to...

32. Molecular analysis of endophytic bacteria from the genus Bacillus isolated from tropical maize (Zea mays L.) - Figueiredo,José Edson Fontes; Gomes,Eliane Aparecida; Guimarães,Claudia Teixeira; Lana,Ubiraci Gomes de Paula; Teixeira,Marta Aparecida; Lima,Guilherme Vitor Corrêa; Bressan,Wellington
Endophytic bacteria play an important role in agriculture by improving plant performance and adaptation against biotic and abiotic stresses. In the present study molecular methods were used for identifying Bacillus endophytic bacteria isolated from Brazilian sweet corn. SDS-PAGE of wholecell protein extract of fortytwo isolates revealed a high number of scrutinable bands. Twenty-four isolates were identified in nine different groups of duplicated bacteria and eighteen were identified as unique. Some high-accumulated polipeptides with variable length were observed in almost isolates. Partial sequencing of 16S ribosomal gene revealed that all isolates are Bacillus sp. and among thirteen isolates with similar protein...

33. ANTIBIOTIC RESISTANT BACTERIA ISOLATED FROM THE AIR OF SWINE CONFINEMENT OPERATIONS - GIBBS, SHAWN G.
The creation of antibiotic resistant bacteria is a global problem that the United States is just now starting to address. The hospital costs of antibiotic resistance are estimated to be hundreds of millions of dollars a year to treat, and they jeopardize the health of the human or animal that is infected. The inhalation of large amounts of airborne microorganisms is also a problem when dealing with those that work with or live in proximity to animal confinement units. It has been shown that the inhalation of large numbers of microorganisms creates a variety of adverse human health effects. The...

35. Direct RAPD evaluation of bacteria without conventional DNA extraction - Araújo,Welington Luiz; Angellis,Derlene Attili de; Azevedo,João Lúcio
The present work reports successful DNA amplification of Pantoea agglomerans and Bacillus pumilus through Random Amplified Polymorphic DNA (RAPD). For this, template DNA was obtained without conventional DNA extraction. The procedure was as follows: cultures grown for 20 hours in 5 mL LB medium were centrifuged and the resulting preparation was suspended in TE buffer. After boiling, the cell suspension was diluted and 2.0 µl were used in reactions of 15 µl. The results showed no significant differences among the RAPD profile of the PCR reactions derived from the boiling and phenol extraction methods, suggesting the utilization of this method...
